چکیده

Abstract Immunity to infections is crucial healthy life. However, recent Covid-19 pandemic has shown that comorbidity such as kidney disease and associated uremia impairs immunity against infections. Uremic patients show high susceptible also exhibit poor antibody responses vaccine. The mechanisms by which negatively impacts response unknown. Using multiple mouse models of uremia, we assessed B cells model antigen NP-KLH in alum. We inhibits both canonical germinal center (GC) non-canonical extra-follicular following immunization. Immunized uremic mice demonstrated compromised affinity maturation, isotype switching, antigen-specific secreting cells, titer T follicular helper response. Consequently, exhibited diminished GC prime-boost showed increased cell cycle arrest, apoptosis impaired migration between light dark zone the GCs. identified toxin hippuric acid a major driver loss mitochondrial membrane potential leading human cells. Finally, were similarly during influenza virus infection, cause mortality with disease. These results shed on how toxin(s) suppress antimicrobial vaccine individuals Knowledge gained from this study may pave path for developing effective therapeutic preventive strategies including SARS-CoV-2. The germinal center antibody response in health and disease

The germinal center response is the delayed but sustained phase of the antibody response that is responsible for producing high-affinity antibodies of the IgG, IgA and/or IgE isotypes. B cells in the germinal center undergo re-iterative cycles of somatic hypermutation of immunoglobulin gene variable regions, clonal expansion, and Darwinian selection for cells expressing higher-affinity antibody...

Germinal center B cells govern their own fate via antibody feedback

Affinity maturation of B cells in germinal centers (GCs) is a process of evolution, involving random mutation of immunoglobulin genes followed by natural selection by T cells. Only B cells that have acquired antigen are able to interact with T cells. Antigen acquisition is dependent on the interaction of B cells with immune complexes inside GCs. It is not clear how efficient selection of B cell...

Cyclooxygenase-1 orchestrates germinal center formation and antibody class-switch via regulation of IL-17.

The cyclooxygenase (COX) enzymes are known modulators of innate immune cell function; however, their contributions to adaptive immunity are relatively unknown. We investigated the roles of COX-1 and COX-2 in the humoral immune response to infection with the Lyme disease pathogen Borrelia burgdorferi. We report that in vitro, murine B cells constitutively expressed COX-1 and up-regulated express...

Germinal Center Selection and the Antibody Response to Influenza

In this Minireview, we discuss basic aspects of germinal center biology in the context of immunity to influenza infection and speculate on how the simultaneous evolutionary races of virus and antibody may impact our efforts to design a universal influenza vaccine.
